L'ONL est composé d'un secrétariat général et d'un comité scientifique dont les dix membres et le président sont nommés par le ministre de l'Éducation nationale pour une durée de trois ans, renouvelable une fois. Les membres sont choisis parmi les enseignants du premier et du second degré, les enseignants-chercheurs, les chercheurs, les membres des corps d'inspection du ministère chargé de l'Éducation nationale, les parents d'élèves ainsi que parmi des personnalités qualifiées.

L'Observatoire national de la lecture est un organe consultatif rattaché au ministère de l'Éducation nationale créé par arrêté du ministre de l'Éducation nationale, de l'Enseignement supérieur et de la Recherche, François Bayrou, le 7 juin 1996.
Il est rattaché, à partir du 15 septembre 1999, à l'Institut national de recherche pédagogique (INRP). Le 3 avril 2001, est créé un nouvel Observatoire, à nouveau rattaché au ministère de l'Éducation nationale. La lutte contre l'illettrisme est supprimée de ses objectifs et remplacée par l'analyse des pratiques pédagogiques. À partir de septembre 2009, s'ajoute à ses missions la réflexion sur les liens entre supports classiques et virtuels de la lecture, ainsi qu'entre patrimoine littéraire et littérature contemporaine. Son fonctionnement est également modifié, le nombre de ses membres passant ainsi de dix-huit à dix.
Le décret nº 2011-1092 du 9 septembre 2011, ainsi que l'arrêté du même jour suppriment l'Observatoire national de la lecture.
L’ONL a pour missions d’analyser les pratiques pédagogiques dans le domaine de l'apprentissage et du perfectionnement de la lecture. Il recueille et exploite les données scientifiques permettant d'éclairer ces pratiques. Il organise, sur ces mêmes sujets, un échange constant d'informations et d'expériences avec les enseignants et les parents d'élèves. Il formule des propositions dans le domaine de la pédagogie de la lecture et de la formation initiale et continue des enseignants. Il lui incombe d’améliorer l'apprentissage et le perfectionnement de la lecture, d’analyser les causes de l'illettrisme chez les jeunes adultes afin de mieux y remédier. Il propose des actions de nature à développer le goût de la lecture chez les jeunes. Il conduit en particulier les réflexions sur les moyens de rapprocher les supports classiques et les supports virtuels de lecture dans le but de maintenir vivace le lien entre les grandes œuvres du patrimoine littéraire national et international et la littérature contemporaine.
L'Observatoire national de la Lecture est un lieu de promotion de recherches, d'études et de formations. C'est également un lieu d'information du public qui met à sa disposition un centre de ressources, relais entre les praticiens et les sources d'informations existantes (données pédagogiques, travaux de recherches, institutions et organismes divers).
Il a publié de nombreux travaux et réalisé chaque année un colloque « Les journées de l'ONL » sur une thématique liée aux questions de la lecture et de son apprentissage.
