Anne-Marie-Louise d’Orléans est la fille unique de Gaston d’Orléans et de Marie de Bourbon. De sa mère, morte six jours après sa naissance, elle hérite de l’immense fortune et des titres de dauphine d’Auvergne, duchesse de Montpensier et de Châtellerault, de princesse d’Orléans, de Dombes et de Joinville, de comtesse d’Eu et de Mortain.
Gaston d’Orléans se préoccupe peu de la fille de Marie de Bourbon, qu’il avait eu tant de répugnance à épouser. La petite fille est donc élevée, sous la houlette de Madame de Saint-Georges, par Louis XIII et Anne d’Autriche, ses parents de substitution. Le remariage de son père avec Marguerite de Lorraine en 1632 n’apporte pas à Anne-Marie-Louise un foyer aimant. La nouvelle duchesse d’Orléans, épousée contre le gré du roi, vit en exil durant dix ans, avant d’obtenir le pardon de son beau-frère, et sa belle-fille la déteste. Anne-Marie-Louise accorde cependant son affection à ses demi-sœurs, Marguerite-Louise, Élisabeth-Marguerite et surtout Françoise-Madeleine, de dix-neuf ans sa cadette, qu’elle considère comme sa fille.
Considérée comme le plus beau parti d’Europe en raison de sa naissance et de sa fortune, la princesse est très imbue de sa personne. Elle exige d’abord de se faire appeler Mademoiselle, comme son père était appelé Monsieur, ce qui la distingue de ses demi-sœurs. Elle deviendra la Grande Mademoiselle, lorsque la naissance de Philippe d’Orléans, le petit Monsieur, fit de son père le Grand Monsieur. Elle se met également en tête d’épouser son cousin Louis, pourtant plus jeune qu’elle de onze ans, et de devenir reine de France. Le refus de Mazarin provoque sa rancœur et la jette dans la Fronde des princes aux côtés de son cousin Condé. Le 27 mars 1652, elle se lance dans une action audacieuse en s’emparant de la ville d’Orléans, persuadée d’être alors en position de force pour réclamer une paix avantageuse pour les princes et surtout pour obtenir d’épouser le petit Louis XIV. Après la bataille de Bléneau (7-8 avril 1652), Condé et la Grande Mademoiselle se replient sur Paris. Assiégé par Turenne dans le faubourg Saint-Antoine, Condé est sauvé par sa cousine qui fait tirer les canons de la Bastille sur les troupes royales. Ces actions d’éclat provoquent la disgrâce de la princesse une fois la paix revenue et, en octobre 1652, elle se voit signifier son exil de la Cour. Elle se replie alors sur son château de Saint-Fargeau et profite de sa solitude pour écrire ses Mémoires, témoignage unique sur la vie de la Cour et la sensibilité d’une femme au XVIIe siècle.
Ayant obtenu son pardon en 1657, elle revient à la Cour, mais en est de nouveau exilée en raison de son refus d’épouser le roi du Portugal, fou et paralytique. Elle ne revient définitivement qu’en 1660, pour assister aux noces de son cousin avec l’infante d’Espagne, ce qui sonne le glas de ses ambitions personnelles.
Ses espérances matrimoniales rebondissent en revanche de manière inattendue en 1669, lorsqu’elle tombe amoureuse d’un gentilhomme d’une naissance bien inférieure à la sienne : Antoine-Nompar de Caumont, marquis de Puyguilhem, comte de Lauzun. En 1670, elle obtient, à force de supplications, l’accord du roi pour ce mariage très inégal. Les courtisans et Marguerite de Lorraine obtiennent toutefois in extremis que celui-ci ne se fasse pas, provoquant la rage des futurs époux. Lauzun insulte alors publiquement Madame de Montespan,dont il sait qu’elle a intrigué contre les noces, ce qui lui vaut dix ans d’emprisonnement à Pignerol. Pour l’en faire sortir, Mademoiselle doit se résoudre à céder sa principauté de Dombes, son comté d’Eu et une partie de ses biens au comte du Maine, fils légitimé de Madame de Montespan. On ne sait exactement si Lauzun épousa réellement la princesse à son retour à la Cour mais celle-ci le comble ouvertement de largesses : le duché de Saint-Fargeau, la baronnie de Thiers et dix mille livres de rentes. Mais l’ingratitude et le mépris qu’il affiche envers elle lui font enfin comprendre qu’il ne l’a jamais aimée. Elle le quitte définitivement en 1684 et consacre ses dernières années à Dieu. Elle meurt le 5 avril 1693.
