Né le 2 septembre 1733 à Montcornet-en-Thiérache (Aisne). Mort guillotiné le 3 février 1794 à Paris.
Son père, Nicolas François Ducloz-Dufresnoy, est directeur des fermes à Toulouse. Sa mère est Louise Regnault (1686-1746). Le partage de la succession de son père a lieu le 11 novembre 1782. Ses soeurs sont Louise Antoinette Ducloz-Dufresnoy (1740-1830) et Marie Jeanne Françoise Ducloz-Dufresnoy (morte en 1794), épouse Marcotte de Pyn.
Il laisse un fils naturel, Charles Athanase Walckenaër, baron Walckenaër (1771-1852), qu'il a eu avec Anne Pajot de Villepenot, marquise Claude Etienne Bidal d'Asfeld. Charles Athanase Walckenaër épouse le 25 mai 1794 sa cousine Joséphine Marie Jeanne Antoinette Marcotte de Pyn, dite "Finotte" (1776-1849), soeur de Charles Marcotte d'Argenteuil et de Philippe Marcotte de Quivières, tous deux collectionneurs et mécènes ; ils ont 3 enfants :
Charles Athanase Marie Walckenaër, baron (1796-1888), marié le 8 octobre 1825, à Rouen, à Albertine Demarest (1807-1840) [lesquels auront également 3 enfants : Marie Charlotte Mathilde W., 1826-1893 ; Charles Marie Guillaume W., 1828-1908 ; Cécile W., 1830-1912].

Le 2 octobre 1787 il emprunte 18000 livres par constitution de 1800 livres de rente viagère.
Syndic de la Compagnie des notaires de Paris en 1789. Il cède son office en 1791 à Louis François Robin (7 juillet 1791, traité d'office). Il exerçait depuis le 6 août 1763, d'abord rue des Déchargeurs (1763-1768), rue Vivienne, près les Filles-Saint-Thomas (1769-1791). Il fut le notaire de Marie Louise O'Murphy, l'une des maîtresses de Louis XV.
En plus d'être notaire au Châtelet, Charles Nicolas Ducloz-Dufresnoy s'intéresse à l'économie. Il fait prêter six millions de livres par la compagnie des notaires à Louis XV. Il tente d'empêcher la création du papier-monnaie et propose un projet pour sauver la Caisse d'escompte. Il publie à Londres, en 1788, L'esprit de M. Necker et, à Paris, en 1789, Origine de la Caisse d'escompte, ses progrès et ses révolutions.
En 1789, il est nommé député suppléant aux Etats généraux et fait partie d'un comité chargé par la Ville d'assurer la sûreté et l'approvisionnement de la capitale. Malgré ou à cause de sa modération, il est guillotiné.
Notaire et mécène du peintre Greuze, il en possède plusieurs tableaux dans sa collection ("La jeune fille en prière", "La dévideuse"), lequel fera son portrait et celui de son fils. Vendue aux enchères, sa collection a fait l'objet d'un catalogue imprimé, dont un exemplaire est conservé à la bibliothèque de l'INHA, collections Jacques Doucet ( Catalogue d'une collection précieuse de tableaux originaux de l'école française, peints par S. Vouet, S. Chardin, Ch. Natoire, J. Vernet, J.-B. Le Prince, J.-B. Greuze, J. H. Fragonard, H. Robert....et autres objets précieux du cabinet de feu le citoyen Duclos-Dufresnoy, par François Léandre Regnault-Delalande, R. Baudoin et Jean Charles Sylvestre, 18 août 1795.
Arrêté sur ordre du Comité de Sûreté générale le 10 nivôse an II (30 décembre 1793), il est condamné à mort par le Tribunal révolutionnaire le 14 pluviôse an II (3 février 1794) et guillotiné le jour même. Une notoriété après décès est établie le 22 prairial an III (10 juin 1795), suivie d'un inventaire après décès le 18 messidor an III (6 juillet 1795). Une mainlevée par ses héritiers et d'autres actes par la suite sont donnés le 29 nivôse an X (19 janvier 1802) [RE/XLVIII/11]. Sa succession est ouverte en février 1793 par son confrère Jean-Baptiste Lefebvre de Saint-Maur.
Sa maison du 118 (auj. 89) Faubourg-Poissonnière est revendue en 1834 par ses héritiers Walckenaër-Marcotte.
[F. Masson] M. Duclos Dufresnoy avait une clientèle fort aristocratique et il fut impliqué au monde du sport si bien que dans les grandes épreuves hippiques, il recevait les paris dans son étude. Il possédait l'une des plus belles maisons de Paris, rue du Faubourg-Poissonnière, et les jardins en furent célèbres au point qu'on venait de loin les visiter. La maison existe encore ; elle est aujourd'hui le Lycée Lamartine. Il avait une nombreuse bibliothèque et une galerie de tableaux composée en majeure partie de l'école française. Il possédait Le triomphe de Bacchus par Natoire, La dame de charité par Greuze et huit têtes charmantes du même, des marines par M. Vernet, ... et plusieurs autres tableaux de MM. Fragonard, Casanova et de Boullogne. La situation de ce notaire était fort enviable et il vivait fort à l'aise en compagnie de sa soeur, Mme Marcotte, restée veuve avec huit enfants. Lui-même ne s'était pas refusé le luxe d'un fils naturel qu'il avait envoyé faire ses études à Oxford et qui fut appelé à de si hautes destinées, comme préfet, littérateur et savant, que le gouvernement en fit un baron et l'Académie des inscriptions, un secrétaire perpétuel.
